[tab] [content title="Summary"] For half a century, it has been established that neurotransmitters are released in preformed quanta, with these quanta representing vesicles that store the neurotransmitter, and that their release occurs via exocytosis. The focus of this book is twofold. The first part delves into the molecular events underlying exocytosis. This includes an analysis of presynaptic calcium channels, the core proteins involved in the secretory machinery, and the effects of clostridial toxins and α-latrotoxin—known for their potency and significant role in advancing our understanding of the exocytotic process. In the second part of the book, the focus shifts to presynaptic receptors for endogenous chemical signals, which regulate neurotransmitter release. These receptors include ligand-gated ion channels and presynaptic G-protein-coupled receptors. The book explores how these presynaptic receptors influence the exocytosis cascade and discusses their therapeutic potential, which is a central theme in most of the chapters.
